ABSTRACT

@#Multifocal electroretinogram(mf-ERG)is a common type of electroretinogram. It can objectively, accurately and quantitatively detect the function of each tiny part of the retina, so it is widely used in clinical, especially in retinal diseases. It not only plays an important role in the diagnosis and efficacy evaluation of retinal diseases, but also has an important value in the follow-up and prevention of diseases, even the scope of its application can be extended to basic research. Therefore, this paper reviews the application of multifocal electroretinogram in retinal diseases.

ABSTRACT

Objective::To analyze and excavate the medication rules of ancient physicians in the treatment of depression syndrome, so as to provide reference for clinical treatment of depression syndrome. Method::The ancient medical records of depression syndrome sorted out in Yian Leiju were retrieved and collated, the data of traditional Chinese medicine (TCM) prescriptions in medical records were extracted, the obtained data of TCMs in prescriptions were standardized, and the data mining function of the ancient and modern medical records cloud platform V1.5 was adopted to carry out frequency statistics, cluster analysis, association analysis and complex network analysis of TCM data, the common drugs used by ancient physicians for treatment of depression syndrome, properties and classifications of commonly used medicines, pairs of commonly used medicines and core prescriptions were obtained. Result::After screening, a total of 364 medical cases, 461 visits and 439 prescription data of TCM were included, involving 67 ancient books. A total of 607 Chinese medicines were involved, and the total frequency of medication was 4 127.High frequency medicines included Poria (186 times), Citri Reticulatae Pericarpium (115 times), Glycyrrhizae Radix et Rhizoma (113 times), Ginseng Radix et Rhizoma (101 times), Angelicae Sinensis Radix (98 times), Atractylodis Macrocephalae Rhizoma (94 times), etc. The main properties and flavors of commonly used Chinese medicines were warm, flat, bitter and acrid, which were mainly attributed to the four meridians of spleen, lung, heart and liver. Commonly used couplet medicines were Atractylodis Macrocephalae Rhizoma-Poria, Poria-Glycyrrhizae Radix et Rhizoma, Poria-Angelicae Sinensis Radix, Atractylodis Macrocephalae Rhizoma-Glycyrrhizae Radix et Rhizoma, Ginseng Radix et Rhizoma-Poria, etc. The core prescription was based on the addition and subtraction of Danzhi Xiaoyaosan. Conclusion::In the treatment of depression syndrome, ancient physicians adopt the principles of invigorating the spleen, nourishing Qi and blood, soothing the liver and regulating Qi, nourishing the heart and tranquilizing the mind, giving consideration to both Qi and blood, and harmonizing the liver, spleen and heart.

ABSTRACT

BACKGROUND: Precartilaginous stem cells exist in articular cartilage, which may become potential seed cells for cartilage tissue engineering. Transforming growth factor-β3 (TGF-β3) has positive regulation effect on the proliferation and chondrogenic differentiation of precartilagious stem cells. OBJECTIVE: To investigate the effect of TGF-β3 on the chondrogenesis of osteochondroprogenitor cells. METHODS: CD146+chondrocytes were isolated from the patients with advanced osteoarthritis, and were then identified. CD146+chondrocytes were cultured in the normal medium (blank control group), chondrogenic induced medium (control group), chondrogenic induced medium containing 2.5 and 10 μg/L recombinant human TGF-β3, respectively. The immunohistochemistry of collagen Ⅱ and aggrecan was performed, and the related gene expression was tested by real-time quantitative PCR after 4 weeks of culture. RESULTS AND CONCLUSION: When chondrogenic differentiation was performed, the number of cell pellets in the 10 μg/L TGF-β3 group was greater than that in the 2.5 μg/L TGF-β3 group, and the number of cell pellets in the 2.5 μg/L TGF-β3 group was greater than that in the control group. The expression levels of collagen Ⅱ and aggrecan in the 10 μg/L TGF-β3 group was significantly higher than that in the 2.5 μg/L TGF-β3 group (P< 0.05). The expression levels of collagen Ⅱ and aggrecan in the 2.5 μg/L TGF-β3 group were significantly higher than those in the control group (P< 0.05). Real-time quantitative PCR results showed that the expression levels of collagen Ⅱ and aggrecan mRNA in the 10 μg/L TGF-β3 group were significantly higher than those in the 2.5 μg/L TGF-β3 group (P< 0.05), but the expression level of SOX-9 showed insignificant difference between two groups (P > 0.05), and the expression level of SOX-9 in the 10 and 2.5 μg/L TGF-β3 groups was significantly higher than that in the control group (P< 0.05); the expression levels of collagen Ⅱ and aggrecan mRNA in the 2.5 μg/L TGF-β3 group were higher than those in the control group (P< 0.05). Our findings suggest that osteochondroprogenitor cells with stem cell characteristics exist in the residual articular cartilage of the patients with advanced osteoarthritis. TGF-β3 has the ability of promoting chondrogenic differentiation of osteochondroprogenitor cells, which may be an ideal cytokine for cartilage tissue engineering.

ABSTRACT

A growing number of children and adolescents are being diagnosed as Chiari malformation type Ⅰ (CM-Ⅰ) for behavioral disorders,developmental delay,seizures,or abnormal orpharyngeal function.The aim of this study was to compare the clinical characteristics,imaging findings and surgical outcomes of CM-Ⅰ in pediatric and adult patients.Between January 2014 and June 2017,84 patients with CM-Ⅰ underwent surgical treatment in our department.We divided the patients into two groups:pediatric group (n=1 1,age <18 years)and adult group (n=73,age ≥18 years).Data on clinical characteristics,imaging findings,surgical outcomes,and prognosis were retrospectively reviewed and compared between these two groups.For clinical presentation,scoliosis (36.4%) and developmental delay (36.4%) were more common in pediatric patients,whereas,sensory disturbance (58.9%) and motor weakness (41.1%) were more common in adult patients.Imaging findings showed that the incidence of hydrocephalus and craniovertebral junctional abnormalities was significantly higher in pediatric group than in adult group (P<0.05).Compared to adult group,pediatric group showed a better improvement or resolution of syrinx and tonsillar herniation after surgical treatments (P<0.05).The total Chicago Chiari Outcome Scale (CCOS) score in pediatric patients at the last follow-up was significantly higher than that in adult patients (P=0.002).In conclusion,the clinical characteristics and imaging findings appeared to be different in pediatric and adult patients with CM-Ⅰ.The surgical outcomes of pediatric patients were shown to be significantly better than those of adult patients.

ABSTRACT

Objective:To investigate the effect of miR-107 targeting NID2 on Notch signaling pathway on invasion and proliferation of lung cancer.Methods: The expression of NID2 in lung cancer and normal tissues was detected by immunohistochemistry.The expression of miR-107 in lung cancer cell line was detected by PCR.The effect of miR-107 on the expression of NID2 was examined by the dual luciferase gene system.Effect of miR-26 on the proliferation ability of H1650 were detected by tumor in ball assays.Transwell invasion assay was used to detect the ability of invasion in the lung cancer cell line A549 after overexpression the miR-107.Scratch test was used to detect the ability of migration in the lung cancer cell line A549 after overex-pression the miR-107.Western blot was used to detect the protein expression of Notch signal pathway after overexpression miR-107.Results:The expression of NID2 in lung cancer tissues was significantly higher than that in normal lung tissues.The expression of miR-107 in lung cancer tissues was significantly lower than that in normal lung tissues.The dual luciferase reporter gene system showed that miR-107 could directly regulate the transcriptional activity of NID2.After miR-107 overexpression,the proliferation,invasion and migration ability of lung cancer cell A549 were significantly decreased.The expression of miR-107 protein could down-regulate the expression of Notch1,hes-1 and presenilin1 protein.Conclusion: miR-107 targets the expression of NID2 and regulates the invasion and proliferation of lung cancer cells by Notch pathway.

Objective: The differences in the compounds between Lysimachia christinae and Desmodium styracifolium were compared in order to provide the references for pharmacodynamic differences of them. Methods: The RP-HPLC-UV and RP-HPLC-ELSD fingerprints of L. christinae and D. styracifolium were established, and the common peaks of two kinds of fingerprints were compared. Results: The HPLC-UV fingerprints of L. christinae were obviously different from the HPLC-ELSD fingerprints, and the strong peaks detected in UV were not the same as what detected in ELSD, so were those of D. styracifolium. There were 14 and 5 common peaks, respectively in the HPLC-UV and HPLC-ELSD fingerprints of L. christinae, while 28 and 18 common peaks, respectively in the HPLC-UV and HPLC-ELSD fingerprints of D. styracifolium. The similarities of chromatograms of D. styracifolium were better than those of L. christinae. Four common peaks were identified in the HPLC-UV fingerprints of L. christinae and D. styracifolium, and two common peaks in their HPLC-ELSD fingerprints. Conclusion: It is better using HPLC-ESLD to establish the fingerprints of L. christinae and D. styracifolium. The chemical constituents of L. christinae were significantly different between the different batches. However, little variation was found in the chromatograms between the different batches of D. styracifolium. The two plants may contain two same compounds with high content, which may have the function as the substantial bases of their treatments for the same indication. However, they may have their special efficacies.

Supported by NSFC molecular pharmacognosy projects and funding a gradual increase in the number of, the proportion of funds for pharmaceutical research funding tends to be stable; mainly supported by molecular biology methods of genuine medicinal materials, secondary metabolism and Germplasm Resources Research; hot drugs including Radix Salviae Miltiorrhizae, Radix Rehmanniae, Cordyceps sinensis, hot contents including tanshinone biosynthesis, Rehmannia glutinosa continuous cropping obstacle.

ABSTRACT

Using the methods of informetrics analysis, articles retrieved from the database of CNKI were statistically analyzed on development course and knowledge system, so as to reflect the overall situation of pharmacognostical studies by molecular biotechnology. The result shows that the research on pharmacognosy by molecular biotechnology is an inter-disciplinary research area, the major research fields can be divided into 7 categories, including molecular identification of Chinese medicinal materials, molecular systematics and genetic diversity analysis of Chinese medicinal materials, biosynthesis and bioregulation of secondary metabolites in medicinal plants, molecular mechanism and genetic basis of Dao-di Herbs, and tissue culture and molecular breeding in medicinal plants. The research on pharmacognosy by molecular have achieved remarkable progress in recent 20 years, and have broad development prospects.

ABSTRACT

The method of bibliometrics was used to analyze the literature about the application of molecular biotechnique to pharmacognosy which were searched and obtained from the CNKI database and Shanghai intellectual property information platform from the year 1995 to 2015.It was found that 22 462 articles were published and the 63% were funded, 50 core institutions and 888 authors, 18 core journals were engaged in this subject.496 items of patents were authorized and 90 kinds of Chinese Materia Medica were involved.In the view of the quantity and quality of published literature, the scale and influence of journals, institutions, and the extent of subject categories have made remarkable achievement. Molecular pharmacognosy has completed the germination stage of a new subject, and has been in a relatively mature and stable development status.

ABSTRACT

Lysimachiae Herba and Desmodii Styracifolii Herba are common traditional Chinese medicines for treating lithiasis. Both of them have efficacies of clearing heat, diuresis and eliminating calculi. However, there are some differences in their clinic applications. The former is mainly used to treat hepatolithiasis, gallstones, jaundice, stranguria and gout; Whereas the latter is mainly used to treat urinary calculus. In this paper, the pharmacological effects of Lysimachiae Herba and Desmodii Styracifolii Herba on removing calculus, choleresis, anti-inflammation and oxidation resistance were compared and analyzed based on document retrieval. In conclusion, both of them show the preventive and therapeutic effects on kidney stones and gallstones. Particularly, Desmodii Styracifolii Herba has a better effect in treating the kidney stones, while Lysimachiae Herba has a better effect in treating cholesterol gallstones.

ABSTRACT

<p><b>OBJECTIVE</b>Study on quality of life of asymptomatic HIV infected persons with traditional Chinese medical, which can provide the clinical basis for improving the quality of life.</p><p><b>METHOD</b>This study applied a randomized, double-blind, and placeb-parallel control designed method to select 1 200 persons in the asymptomatic period of HIV infection as the subjects. The subjects were randomly divided into the treatment group and the control group at the ratio of about 2:1. According to the results of monthly differential diagnosis of TCM, the test group and the control group were given homologue Chinese drugs preparations and model Chinese drugs. The total study period was 18 months. Using PRO scale and the world health organization AIDS determination of quality of life short scale form (WHOQOL-HIV-BREF) to investigate asymptomatic HIV infected persons, according to different times, we calculated the total score and each domain score of quality of life of the treatment group and control group, we did statistical analysis.</p><p><b>RESULT</b>Form the PRO scale,we can see that the treatment group showed a trend of stability, compared with the control group with significant statistical difference (P < 0.05) after 6 months; from the WHOQOL-HIV scale analysis, we can see that compared with before treatment, the quality of life of the treatment group was increased, the difference was significant (P < 0.05), but the quality of life of the control quality of life was decreased, the differences was significant (P < 0.05).</p><p><b>CONCLUSION</b>Dialectical therapy of Chinese medicine can significantly improve the patient's quality of life, which can provide the basis for the prevention and control policy formulation and implementation with asymptomatic HIV infected persons.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the gene polymorphisms of ApoAI-75 Msp1, ApoB Msp1, ApoCIII Sst1, LRP5, and ApoE genotypes in two pairs of semi different modes of hepatitis B for HBV markers.</p><p><b>METHODS</b>The patients are divided into 9 groups. There were a total of 720 cases, 80 patients in each group, The patients was carried out by SnaPshot method (single-base multilocus micro-sequencing), and different genotypes of each locus were conducted by the method of sequencing in order to support the final evidence of the accuracy of test results.</p><p><b>RESULTS</b>There was association between gene polymorphisms of ApoAI-75Msp1 and ApoE and different modes of two pairs of semi-hepatitis B (P < 0.05), while there wasn't any association between gene polymorphisms of ApoB-Msp1, ApoCIII-Sst1, LRP5 and different modes of two pairs of semi-hepatitis B (P > 0.05).</p><p><b>CONCLUSION</b>The gene polymorphism of ApoAI-75Msp1 and ApoE was associated with the different modes of HBV markers.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the clinical effect of the treatment of the ununited lower limb fracture with reamed intramedullary interlocking nail.</p><p><b>METHODS</b>Fifty-two patients with ununited lower limb fracture were treated with intramedullary interlocking nail (femur 36, tibia 16). The main cause of nonunion was interternal fixation loosening or fracture (41 cases, 78.8%), other reasons included bone defection, bone infection or too much traction of fracture (11 cases, 21.2%). According to X-ray classification: 41 cases in type hypertrophy, 11 cases in type atrophy. All these patients need operation to remove the primary implant, clean the hard bone, ischemia bone and scar, manual ream and fix by intramedullary interlocking nail, then bone graft were done around the fracture site. With the knee joint stiffness, relax it by manipulate or operation.</p><p><b>RESULTS</b>Beside 3 superficial infection (2 delayed healing and 1 healing after debridement), other 49 wounds healed on time. All cases were followed up for an average of 25.2 months, ranging from 12 months to 39 months. All the ununion healed from 5 to 11 months, mean union time were 6.1 months. Deep tissue infection, fat embolism, implant loosening, nail broken or nonunion were not found in this series. According to judet standard on the movement of knee, the excellent-good rate was 82.7%.</p><p><b>CONCLUSIONS</b>Intramedullary interlocking nail is effective to treat nonunion of lower limb fracture.</p>

ABSTRACT

<p><b>BACKGROUND</b>To study arboviruses carried by mosquitoes in Liaoning Province in 2002.</p><p><b>METHODS</b>Totally 4927 mosquitoes were collected from Liaoning Province in July 2002. Virus strains were isolated by inoculating BHK-21, C6/36 and Vero cells. The newly isolated strains were identified by serological (ELISA and IFA) and molecular methods (Real-Time PCR and RT-PCR).</p><p><b>RESULTS</b>Two strains were isolated from mosquitoes causing cytopathogenic effect (CPE) on cells and were fatal to suckling mice. Serological tests showed that both were positive for the antibody to JEV. The PrM and E gene were cloned and sequenced. The phylogenetic analysis showed that the new isolates belonged to genotype I, JEV. Sequence analysis showed that the homology of nucleotide sequences and amino acid (AA) sequences between the two strains was 100%. Compared with the nucleotide sequences between the two strains and the standard JEV vaccine strain SA14-14-2, the difference was up to 4.11%, and the difference of AA was 0.6%.</p><p><b>CONCLUSION</b>Two strains of JEV were isolated and identified in Liaoning province, both belonged to genotype I JEV.</p>

ABSTRACT

<p><b>BACKGROUND</b>Mosquitoes were collected in Heilongjiang province in 2002, four virus strains were isolated by inoculation of homogenates onto BHK cell lines. The viruses were identified. Multiple alignment and phylogenetic analysis were carried out by Clustal X (1.8) program.Amino acid (AA) analysis was carried out by GENEDOS (3.2).</p><p><b>RESULTS</b>Biological characters of four newly isolated strains were examined and it was found that all of them could produce cytopathogenic effect (CPE) in BHK cells, killing sucking mice. Serological tests showed that all of these stains reacted positively to JEV antibodies. PrM and E gene regions were amplified and sequenced. Phylogenic analysis showed that all the newly isolated JEV strains belong to genotype III. Using the vaccine strains (SA14-14-2) as control, analysis of the E gene of the new strains and two JEV strains (47, Ha-3) isolated previously from Heilongjiang province showed that these new strains' nucleotide sequence had a homology of up to 99.9% and the amino acid sequence homology up to 99.8%, respectively. Compared with the standard JE vaccine strain SA-14-14-2 and the four new strains, the nucleotide sequence homology was 97.3% and amino acid sequence homology was between 96.8% and 97.0%, respectively. Compared with vaccine strain, there were seven common variations in all the four newly isolated strains.</p><p><b>CONCLUSION</b>Four JE virus strains were isolated in Heilongjiang province. As compared to the vaccine strain, six variations were found in the newly isolated strains at the eight sites relevant to the virulence of the virus.</p>

ABSTRACT

Objective To investigate changes of serum cystatin C(Cys C),uric acid,HsCRP, IL-6,TNF-?,and lipids in patients with primary and secondary type Ⅱ b hyperhpidemia.Methods Seventy- five patients with primary and secondary type Ⅱb hyperlipidemia and 75 healthy controls were included in this study.Serum levels of TC,TG,HDL-C,LDL-C,apoAI,apoB,Lp(a),Cys C,UA,HsCRP were measured by immunoturbidimetric assay on AU-640,and IL-6,TNF-? were detected.Results Patients showed higher serum TC,TG,LDL-C,apoB,UA,Cys C and HsCRP than the controls.The levels in patients were(3.70? 0.66)mmol/L,(9.50?0.92)mmol/L,(4.50?0.31)mmol/L,(1.70?0.21)g/L,(1.21?0.17) mg/L,(441.4?114.7)?mol/L,(11.5?3.2)mg/L respectively,while levels in controls were(1.88? 0.66)mmol/L,(4.00?0.66)mmol/L,(2.80?0.33)mmol/L,(0.74?0.23)g/L,(0.71?0.18) mg/L,(343.0?113.9)?mol/L,(1.8?0.7)g/L respectively.Serum Cys C was positively correlated with HsCRP and UA(P
